Homozygous genome editing was used to fluorescently tag subunits of Condensin I and II in HeLa cells. Absolute abundance, spacing, and dynamic localization of the tagged proteins during mitosis was recorded by fluorescence correlation spectroscopy (FSC)-calibrated live-cell imaging and superresolution microscopy.
